The specific mechanism cited by reviewers: MUS (Eq. 11) is built from spatial contiguity and neighborhood-consistency terms that the spatial-regularization loss (Eq. 9) directly optimizes, so a MUS/contiguity rise at M4–M5 cannot currently be distinguished from the spatial loss dominating as noisier features are added, versus genuine biological signal. No permutation test on chromatin blocks, no independent ground truth (ChIP-seq, motif enrichment), no MUS-with-λ₃=0 ablation is provided.
